What are the molecular mechanisms whereby the liver transports metabolites, drugs, substrates, lipids, and proteins from blood into the bile? How does the hepatocyte, which is a polarized cell, restrict these functions to specific domains of the plasma membrane? How do the specific transporters (which we have identified) traffic intracellularly? How is their function(s) regulated? Answers to these questions are essential for understanding many acquired, inheritable, and developmental defects in transporter function which we have identified in patients and animals with liver disease.
