I am a molecular pharmacologist/endocrinologist with a primary interest in G-protein coupled receptors. These seven transmembrane domain proteins reside on the cell surface and are considered a major target of current drug discovery efforts. Research in the my laboratory focuses on the mechanisms that enable high-affinity ligand-receptor interactions, as well as ligand-induced stimulation of intracellular signaling. As model systems to explore these processes, the specific receptors for two distinct types of peptide hormones are being studied, (i) cholecystokinin/gastrin (which are involved in multiple functions both in the gastrointestinal tract and in the central nervous system) and (ii) glucagon-like peptides (incretins/neurotransmitters that play an important physiological role in regulating blood glucose homeostasis, intestinal nutrient resorption, and food intake). Among these, the receptor for glucagon-like peptide-1 is of particular interest given its potential for the drug or gene therapy of diabetes and obesity.
