We use X-ray crystallography in conjunction
with other biochemical and biophysical techniques to study the
structure and function of the roughly one dozen proteins which
together constitute the yeast cleavage/polyadenylation complex.
Similar macromolecular assemblies are present in all eukaryotic
organisms, and serve to truncate and polyadenylate the 3' end
of messenger RNA prior to nuclear export. We are particularly
interested in understanding the structural basis for the processivity
exhibited by poly(A) polymerase, the enzyme at the core of this
complex. We are also using computational docking and structure
based drug design methods to identify small molecule inhibitors
of the anthrax toxin, Edema Factor.
