Our research is about cellular aging. Protein degradation rates decline with age, and most of the decline is due to reduced activity of a lysosomal pathway of proteolysis that is selective for particular cytosolic proteins. In this proteolytic pathway, protein substrates are targeted to the lysosome after interacting with a molecular chaperone (hsc73) in the cytosol. The substrate protein then interacts with a receptor in the lysosomal membrane, and is next pulled into the lysosome by the actions of a lysosomal form of hsc73. This pathway normally is activated during prolonged starvation. The cause of the age-related decline in this proteolytic pathway is a reduced amount of receptor in the lysosomal membrane.
