Our laboratory studies the signaling of protease-activated receptors (PARs). Thrombin cleavage of the thrombin receptors activates a diverse array of physiologic responses, including platelet aggregation, cellular proliferation/apoptosis, and potentially the invasive and tissue-reorganizing processes involved in cancer. Activation of platelet thrombin receptors may play a major role in the initiation and maintenance of pathological arterial and venous thromboses and the development of atherosclerotic lesions. We study the mechanism of thrombin receptor activation, G protein coupling, downstream signal transduction, and signal shut-off. Projects include: 1) role of PARs in cancer cell invasion and cell motility, 2) thrombin-receptor interactions and development of novel anti-PAR therapeutics, and 3) receptor-G protein signaling in platelets and cancer cells.
