Our laboratory has previously demonstrated that T cell costimulation through VLA-4 integrin ligation inhibits the centralization of SLP-76-containing signaling complexes. My goals are 1) to identify specific mechanisms by which integrin ligation immobilizes SLP-76 microclusters, and 2) to determine whether the integrin-dependent immobilization of SLP-76 microclusters is sufficient for optimal T cell costimulation, or whether the tension generated by resisting the movement of SLP-76 also plays a role in T cell costimulation. To address these questions, I am selectively perturbing integrin-dependent signaling pathways, and examining T cell function and microcluster movement in response integrin ligation. |
